Outline of Annual Research Achievements

本研究では、RNA 修飾の変動と制御という新しい概念を確立し、エピトランスクリプトミクス研究におけるパラダイムシフトを目指す。今年度は新規RNA修飾酵素の同定と機能解析で大きな進展があった。
5-カルボキシメトキシウリジン(cmo5U)は、遺伝暗号の解読に重要な役割を担うtRNA修飾である。私たちは、比較ゲノム解析とRNAの質量分析法を駆使した解析により、cmo5U生合成の最初の水酸化反応を担う二つの遺伝子trhPとtrhOを発見した。TrhPはプレフェン酸依存的なtRNAの水酸化酵素であり、TrhOは空気中の酸素を用いてtRNAを直接水酸化する酵素であった。この研究で、cmo5U修飾は、細胞内におけるシキミ酸の代謝経路および環境中の酸素の有無を感知し、細胞がおかれた多様な環境に適応するための機構であることが示唆された(Nature Commun., 2019a)。
3-(3-アミノ-3-カルボキシプロピル)ウリジン(acp3U)は、バクテリアから真核生物まで、様々な生物のtRNAに存在するRNA修飾である。私たちは、比較ゲノムにより候補遺伝子を絞り込み、RNA-MSを用いた解析により、tRNA aminocarboxypropyltransferase(TapT)と命名した新規酵素を発見した(Nature Commun., 2019b)。acp3U修飾にはtRNAに熱安定性を付与する機能があることや、tapT遺伝子を欠損すると継続的な熱ストレス下でゲノムに変異が生じ、細胞の運動性に異常が顕れることを見出した。さらにTapTのヒトホモログであるDTWD1とDTWD2を見出した。いずれもヒトの疾患との関係が報告されていることから、今後は、acp3U修飾が担う生理学的な役割についての解明が期待される。

1: Research has progressed more than it was originally planned.

Reason

新規RNA修飾酵素の同定とその機能解析では、予想以上の成果が得られた。特に長年その生合成経路が不明であったcmo5U修飾の最初の水酸化修飾に関わる二つの遺伝子trhPとtrhOを発見した成果(Nature Commun., 2019a)は、大きな反響があり、問い合わせや研究試料の依頼が相次いだ。さらに、acp3U修飾酵素であるTapTの同定(Nature Commun., 2019b)は、最終的に私たちを含め、3グループが競合したが、私たちが一番先に論文を発表することができた。生合成機構だけではなく、acp3U修飾の機能や欠損株の表現型を明らかにした点、ヒトホモログであるDTWD1とDTWD2を見出した成果も加わり、他の二つのグループが報告した内容とは一線を画すものであった。

Strategy for Future Research Activity

RNA修飾が生育環境や細胞内メタボライトによってダイナミックに変動する現象を引き続き探求する。特に嫌気環境におけるRNA修飾の変動と役割について集中した研究を行う予定である。新規RNA修飾については、現時点で、未発表のものを10種類以上発見している。この中には、新規キャップ構造、RNAリン酸化修飾、欠損型塩基など、これまでに類の見ないタイプのものが含まれている。本研究では、新規RNA修飾の更なる探索を継続すると共に、規修飾の化学構造の決定、生合成機構の解明、機能解析を行う予定である。
