Outline of Annual Research Achievements

鬱病は心血管病発症の独立した危険因子であり、心筋梗塞後の予後にも大きく影響している。しかしながら、鬱病における血栓形成増大機序については殆ど明らかにされていない。申請者らは、野生型マウスを用いて鬱様行動を示すモデルマウスの作製に成功した。作製したマウスの頸動脈にFeCl3を塗布し血栓形成モデルを作成した。形成された血栓容積は対照マウスと同等であったが、血栓内のフィブリノーゲン陽性領域は有意に増大していた。血栓病変内における好中球の集積および細胞外トラップ(NETs)形成は鬱様行動を示すマウスにおいて有意に増大していた。さらに、DNase Iを投与したところ、鬱様行動を示すマウスにおける血栓内のフィブリノーゲン陽性領域、好中球集積、およびNETs形成は対照マウスと同等まで抑制された。フィブリン血栓形成には、好中球と活性化血小板との相互作用が深く関与していることが報告されている。各種刺激に対する血小板凝集能および好中球NETs形成能について細胞レベルでの解析を実施した。鬱様行動を示すモデルマウスにおける好中球は、活性型血小板添加により好中球細胞外トラップを起こしやすいことが示された。
