Outline of Research at the Start

遺伝子改変ラットを用いた青班核ノルアドレナリンニューロンからのドーパミンシグナル伝達機構及び新奇体験による記憶保持の強化の分子機構の解明を目的とする国際共同研究をデンマークAarhus大学DANDRITE研究所の竹内倫徳博士と共に計画した。基課題にて開発されたコンディショナル低分子タグ発現カセットを用いたドーパミン受容体、ドーパミン生合成酵素等を標的としたノックインラットを作製して標的タンパクの発現、動態解析と行動試験を竹内研究室で行い、基課題の研究の発展性を実証しつつ本課題の目的を達成する。また、記憶保持の強化過程の解析に有用なcFos-tTAノックインラット系統の解析も行う。

Outline of Final Research Achievements

The principal investigator planned an international collaborative research project to elucidate the molecular mechanisms underlying dopamine signaling from the locus coeruleus, as well as the enhancement of memory retention through novel experiences, using genetically modified rats.
As part of this study, we developed an innovative genome editing technology called the AAV-GONAD method and generated several genetically modified rats that are useful for analyzing dopamine signaling and learning and memory. Additionally, we successfully established a new rat ES cell line and developed new developmental engineering techniques for producing genetically modified rats. While the ultimate goal was not achieved, significant related results were obtained.

Academic Significance and Societal Importance of the Research Achievements

本研究により開発されたAAV-GONAD法はラットだけでなくマウス(および他の動物種)にも適用可能な長鎖ノックイン動物作製技術であるため、生命科学研究の発展について非常に重要な学術的意義を有している。また、神経科学研究に有用な各種の遺伝子改変ラット作製に伴い、新規ラットES細胞の樹立や、胚盤胞補完法によりマウス体内でラット精子を作り出すという発生工学的な新技術の開発にも成功したことから、複雑な遺伝子改変ラットを低コスト、かつ容易に樹立できる環境を整備したと言える。さらにこれらは生殖医療などにも応用できる可能性のある技術であり、社会的意義も大きいと思われる。
