Research Abstract

〈背景〉昨年度は、SIGNR1がTLR2およびDectin-1と共同し酵母に対する細胞応答を昂進することを報告した。今年度はSIGNR1による日和見感染菌であるCandida albicans (C. albicans)生菌の認識機構を検討した。〈結果〉可溶性四量体SIGNR1およびDectin-1を作製し、前者のみが生菌C. albicansの酵母型並びに菌糸型の双方に結合する事を確認した。さらにSIGNR1/Dectin-1共発現RAW264.7細胞にC. albicansを感染させたところ、SIGNR1のみが菌体周囲に局在する事を確認した。更に、SIGNR1がRAW264.7の生菌C. albicans刺激後のTNF-α産生を昂進することも確かめた。次に、SIGNR1の生菌C. albicans上のリガンドを同定するために、始めにC. albicans J-1012(血清型A)由来精製マンノプロテインが、可溶性SIGNR1の菌体への結合が阻害すること、および固相化同マンノプロテインに可溶性SIGNR1が結合する事を示した。さらにマンノプロテイン中のN-グリカンの側鎖構造が異なるCandida属(C. albicans NIH B-792、C. parapsilosis、C. lusitaniae)、および野生型並びに遺伝子欠損S. cerevisea((mnn2)(mnn1, mnn4))菌株由来マンノプロテインを用いて各種阻害実験を行い、SIGNR1はN-グリカン中のα1,2-マンナン側鎖を認識する事が明らかにした。更にSIGNR1はα-マンノシダーゼ処理により作製した非還元末端にβ-マンナンを有するa1,2-マンナン鎖にも結合した。このことから、SIGNR1は非還元末端だけでなく側鎖内部のα1,2-マンナンをも認識する特異なレクチンであることが明らかになった。
