Research Abstract

糖質加水分解酵素は、その反応機構の違いから「アノマー保持型」と「アノマー反転型」の2種に大別されるが、オリゴ糖合成に使用できるのは「アノマー保持型酵素」に限られるとされてきた。もし「アノマー反転型酵素」をオリゴ糖合成のツールとして利用できるようになれば、その応用範囲は飛躍的に広がることとなる。本課題においては、1,2-α-L-フコシダーゼ、還元末端オリゴキシラナーゼ、β-キシロシダーゼなどの「アノマー反転型酵素」を材料にして、その機能改変に取り組み、新規なオリゴ糖合成酵素へと変換することに成功した。
