Research Abstract

AMPキナーゼ(AMPK)は、細胞内AMP濃度上昇によって活性化し、グルコースや脂肪の利用、遺伝子発現、蛋白合成を調節してATPを回復させる酵素であり、"metabolic sensor"、"fuelgauge"として知られている。視床下部室傍核は古くから摂食行動調節に重要な神経核として知られている。そこで本研究では、レンチウイルスを用いて活性型AMPKをマウス視床下部室傍核の神経細胞に選択的・恒常的に発現させ、摂食量と体重の変化を調べた。その結果、室傍核に活性型AMPKを発現させたマウスは過食となり肥満することが判った。興味深いことにこのマウスは、炭水化物食を好み高脂肪食を嫌うことを見出した。さらにこのような食餌嗜好性の変化が、AMPKによる室傍核での脂肪酸酸化の変化によることを見出した。
次に、高脂肪食と炭水化物食に対する嗜好性が肥満動物ではどのように変化しているのか、また、室傍核AMPKとどのような関連にあるかを調べた。その結果、高脂肪食による肥満、KKAy肥満マウスでは脂肪に対する嗜好性が亢進することを見出した。さらに、室傍核AMPK活性及び脂肪酸酸化が低下していた。これらの実験結果は、室傍核AMPK-脂肪酸酸化機構が肥満動物の食餌嗜好性にも調節作用を営むことを示唆する。
