Research Abstract

ヒトに存在するペプチドホルモン、エンドセリン(ET-1)は、血管収縮ペプチドとして発見された。ヒトとげっ歯類ではペプチド3種(ET-1, ET-2, ET-3) が遺伝子ファミリーを形成し、受容体2種と共にET ペプチドホルモン調節系を構成し、系として多彩な機能を担っている。ET ファミリーに関する論文は多数あるが、げっ歯類以前ではET 調節系の詳細は、ヒトET の下等動物に対する薬理的効果を除き、ほとんど未解明である。そこで本研究では、ET 調節系の遺伝子群の起原や分子進化を解明する。動物界における血管収縮ペプチドET ファミリー3種の分布と配列・構造の多様性を解析するために、各種動物(哺乳類から魚類まで)の遺伝子・蛋白質データベースから関連・類似配列を探索した。データベースがないあるいは探索できない動物に関しては、遺伝子クローニングを実施した。塩基配列解析からペプチド配列を明らかにし、前駆体蛋白質の全長を明らかにした。NJ 法(解析ソフト)等で、哺乳類から魚類に至るペプチド前駆体遺伝子の分子進化を解析した。
