Research Abstract

私たちはα1Aアドレナリン受容体(以下α1A受容体)の相互作用分子として細胞内小分子であるスナピンを同定し、その生体内における機能的意義を解明する目的で研究しています。私たちはこれまでに、スナピンがα1A受容体のみならずTRPC6 Ca^(2+)チャネルとも相互作用し、この受容体とチャネル間に生理的リンクを形成する重要な介在分子であること、その結果としてα1A受容体作動性のTRPC6 Ca^(2+)チャネルを介するCa^(2+)流入機構において中心的な役割を担っていることを明らかにしそのメカニズムを報告してきました。
今回私たちはこの受容体作動性Ca^(2+)流入機構が生体内で担う機能的役割の一つとして神経伝達物質の遊離に着目し、PC12細胞を用いてドパミン分泌への関与を調べました。その結果、スナピンを強発現したPC12細胞ではα1A受容体の活性化によりCa^(2+)流入が著しく増大すると共に、細胞外へのドパミン分泌が増大することが観察されました。さらにこのようなドパミン分泌量の増大は1) α1A受容体刺激後の後期相で観察される受容体作動性Ca^(2+)流入の増大に依存する成分と、2) 受容体刺激直後に観察されるスナピンとSNARE複合体間の相互作用によると考えられる成分の2種類から成っていることが示唆されました。
