Research Abstract

腎組織障害時に腎固有の修復・再生に関わる機構が惹起されていないかを検討した。腎障害後の腎糸球体病変および腎尿細管病変を選択的に修復する新規ガンマラクトン化合物(#523と#1376)を2種類のラット腎障害モデル、puromycin腎障害モデルと一側尿管閉塞解除(UUO-release)モデルに投与し、細胞の生存に関わる分子であるheterodimeric aminoacid transporter (HAT)のheavy chain である4F2hc(CD98)とrBATの障害腎における局在を検討した。#523とrat rIFN-γ投与群は糸球体障害を軽減し、4F2hcを発現する単核球細胞が、小葉間動脈周囲リンパ組織、糸球体内毛細血管内、ボーマン嚢上皮、糸球体周囲ボーマン嚢に接して局在した。#1376とrat rTNF-alpha治療群では解除7日目のs-Cr値が1.22mg/dlと対照群の2.28mg/dlより優位な回復を示し、治療群の修復尿細管と尿細管周囲毛細血管内皮細胞にrBATの強い発現がみられた。小葉間動脈周囲リンパ組織と尿細管周囲毛細血管内皮細胞は腎障害後の修復機構である可能性が示唆された。
