P2X receptor-mediated modulation of nociceptive transmission in neuropathic pain model rats

Research Abstract

ATPP2X受容体は末梢における痛覚受容に関与するだけでなく、脊髄後角においても侵害感覚情報を増強することが明らかとなり、大変注目されている。今回、神経因性痺痛モデルラット脊髄スライス標本を用いたパッチクランプ記録を行い、脊髄後角深層細胞のシナプス後細胞におけるATP受容体の役割を検討した。代謝安定型のATP受容体広作動域作動薬であるATP-γSを潜流投与すると、約40%の脊髄後角深層細胞において内向き電流と興奮性シナプス後電流の発生頻度の増強効果が観察された。一方、α, β-methylene ATPの潜流投与を行うと内向き電流は観察されず、興奮性シナプス後電流の発生頻度の増強効果のみが観察された。ATP-γSにより生じた内向き電流は、各種P2Y受容体作動薬の潜流投与によって再現されなかった。また、細胞膜G蛋白質阻害薬存在下において、ATP-γS潜流投与による内向き電流は抑制されなかった。さらに、ATP-γS潜流投与によって生じた内向き電流ならびに興奮性シナプス後電流の発生頻度の増強効果は、P2X受容体括抗薬であるPPADS(10μM)の存在下において完全に阻害されたが、TNP-ATP(20μM)の存在下において影響されなかった。以上の結果から、ATP P2X受容体は脊髄後角深層細胞のシナプス前のみならず、シナプス後細胞においても発現しており、シナプス前とシナプス後細胞に発現しているP2X受容体は異なるサブタイプであることが明らかとなった。このように様々なサブタイプのP2X受容体が広く脊髄後角に分布しており、P2X受容体は末梢からの感覚情報を多彩に修飾していることが示唆された。
