Research Abstract

昨年度は,石灰化過程ならびにコラーゲン線維の走行を観察する方法を確立し,倒立型顕微鏡ステージ上に設置可能な培養装置を試作するとともに,鶏雛脛骨を厚さ200μmに薄切して無菌的に培養する方法の確立に成功した.そこで研究後半の本年度は,骨薄切試料に力学負荷が可能なように培養装置を改良するとともに,準静的ならびに動的な負荷を加えつつ骨試料を培養し,骨形成過程に与える力学負荷の影響を調べた.すなわち,鶏雛脛骨の骨軸方向に垂直な薄切試料の両端をバネ機構を利用してクランプし,試料面内方向に引張・圧縮を加えることのできる装置を試作した.予備的試験の結果,試料に引張を安定して加えることは可能であったが,圧縮は試料の座屈により加えることが困難であった.この装置を用いて試料に準静的な引張を24時間加えたところ,試料に加わる引張ひずみが5%以上の場合,石灰化が有意に促進されることが明かとなった.ひずみが3%以下の場合には静置培養された試料と有意差がなかった.また,石灰化の方向を調べたところ,試料の引張方向に石灰加速度が有意に早いことが明かとなった.次に,試料の引張に伴うコラーゲン線維の走行変化を偏光顕微鏡を用いて調べたところ,石灰化の進行方向はコラーゲン線維の走行方向とよい相関があること,石灰化の速度が上昇するひずみ3%前後を境として,コラーゲン線維の走行が引張り方向に揃うことなどが明らかとなった.コラーゲンの配向が幼若骨組織の石灰化を促進・誘導する可能性が示唆された.
