Research Abstract

カスパーゼは細胞死のメディエーターとして機能するシステインプロテアーゼであり、生体防御機構においても重要な役割を担うことが知られている。そこで、カスパーゼの活性化を可視化するツールとして、FRETを用いたインディケーター、SCAT3(Sensor for activated CAspases based on FRET)を発現するショウジョウバエ系統を作製した。ショウジョウバエの脂肪体は感染時に抗菌ペプチドを産生する器官として知られている。我々は、ショウジョウバエの遺伝学的手法(GAL4/UASシステム)を用いてショウジョウバエの脂肪体にSCAT3を発現させ、感染後のカスパーゼ活性検出を試みた。ショウジョウバエ腹部にE.coliをマイクロインジェクションした後に、時間経過毎に脂肪体を取り出し、単一細胞レペルでのカメパーゼ活性を観察した。その結果、E_coliの注入後30分の間において、脂肪体細胞の一部の細胞で、細胞死を誘導するのに十分なカスパーゼ活性が観察された。感染後の免疫担当細胞におけるカスパーゼ活性化動態の単一細胞レベルでの観察はこれまでに報告が無く、新しい知見である。カスパーゼ活性が観察されたのは一部の細胞だけではなく、それ以外の細胞においても、微弱な、あるいは一時的なカスパーゼ活性が検出された。この活性が、持続し細胞死を誘導するものかどうかについて、E_coli注入後6時間の脂肪体細胞を観察したところ、活性化が見られなかったことから、感染後初期に誘導された一時的な活性化であることが考えられ、E_coliの感染初期においてカスパーゼが何らかの生理機能を果たしていることが示唆された。
