エピジェネティックに失活した癌抑制遺伝子を標的とした新規癌予防法の開発

Research Abstract

発癌の普遍的な機構として、DNA塩基配列の変異を伴わずに、後成的な修飾により起こるエピジェネティックな遺伝子発現の変化が注目され、遺伝子プロモーター領域のDNA高メチル化や、ヒストンの脱アセチル化による遺伝子発現減少等が、発癌に深く関与していると考えられている。このような、エピジェネティックな要因で消失した癌抑制遺伝子の発現を回復できる薬剤や食品成分を発見し、その作用機構を解明できれば、画期的な癌の分子標的予防法の確立に繋がると考えられる。現在までのところ、癌抑制遺伝子p16が高メチル化により失活したヒト癌細胞において、細胞増殖抑制効果のみられた食品成分中に、p16の発現回復がみられるものは見つかっていない。これに対し、昨年、p16が失活したヒト癌細胞において、p16と機能的に同一と考えられるp15が、ゲフィチニブ等によりMEK経路を阻害すると誘導され、p16の機能を代償できることを報告した。現在のところ、MEK阻害剤の感受性は細胞間で大きく異なり、耐性因子等不明な点が多い。MEK阻害剤耐性克服法の確立を目指し、某企業との共同研究により計画を進行中である。またヒストン脱アセチル化については、昨年新規ヒストン脱アセチル化酵素阻害剤(HDACI)YM753が、癌抑制遺伝子p21の発現を誘導すること、マウスモデルでも抗癌効果を示すことを報告した。本年は、p16が高メチル化により失活した細胞を含む種々のヒト癌細胞において、バイカレイン、ケンフェロール等の天然成分や、アロプリノールやジピリダモール等の薬剤が、これまで当教室でHDACIにより発現誘導されることを確認しているDR5遺伝子の発現誘導を介して、抗腫瘍性サイトカインTRAILの感受性を増強し、効果的に癌細胞の細胞死を引き起こすことを見出し、報告した。これらの物質がHDACに対しどのような作用を示すのか等、今後さらに発癌や癌抑制に関わる遺伝子のエピジェネティクスについて研究を継続する。
