Research Abstract

加齢黄斑変性、網膜色素変性、および網膜剥離では、網膜の光受容細胞である視細胞が選択的に障害される。唯一の光受容細胞である視細胞が変性、消失するとその他の細胞が正常であっても、失明に至る。このような失明に対する視機能再建方法として、世界各国で人工網膜が研究されている、人工網膜には、埋め込み部位によって網膜上、網膜下、経強膜など様々なタイプが存在するが、そのいずれもが、出力細胞として残存する網膜神経節を利用して脳への情報伝達を行っており、視細胞変性後の神経節細胞の機能を知ることは重要である。昨年度、我々は生後6ヶ月齢RCSラットおよび生後10ヶ月齢のRCSラットでは生後10ヶ月齢RCSラットで、神経節細胞数が顕著に減少していることを報告した。本年度は、機能的な評価を行うとともにチャネルロドプシンを導入したラットで神経節細胞数の減少が抑制されるかどうかについて検証した。生後6ヶ月のRCSラットの網膜神経節細胞数は約1600個/mmで遺伝子導入後10ヶ月(生後16ヶ月)で1400個/mm、チャネルロドプシンを導入しないラットでは、800個/mmであった。このようにチャネルロドプシンの導入によって、顕著な生存保護効果が認められた。この作用機序を調べる目的で、生存に関わる因子について免疫組織化学により調べた。ChR2を遺伝子導入をしたRCSラットでは、 NF-kBの免疫反応性が神経節細胞層、内網状層で強く認められたのに対し、遺伝子導入していないRCSラット(生後16ヶ月)では免疫反応性が顕著に低下していた。
以上の結果から、ChR2発現による神経節細胞保護には、NF-kBの発現が関与している可能性が示された。
