Research Abstract

I.Aurora A キナーゼが関わる微小管形成経路の解明われわれは、線虫初期胚にγ-チューブリンとAuroraAキナーゼがそれぞれ独立に寄与する微小管合成経路が存在することを見いだしている。われわれは線虫AuroraAキナーゼの活性化型(リン酸化型)のみを特異的に認識する抗体を作成し、Aurora A蛋白質は中心体と微小管上に検出されるのに対して、活性化型Aurora Aキナーゼは中心体の中央部に特異的に局在することを見いだした。さらに、変異型(不活化型)Aurora Aを用いたin vivo解析により、Aurora Aのキナーゼ活性は中心体成熟には必須だが、微小管合成には必要ではないことを見いだした。以上の結果から、Aurora Aのキナーゼ活性依存的機能に加えて、キナーゼ活性非依存的機能が紡錘体形成に重要であることがはじめて明らかとなった。
II.線虫γ-チューブリン結合タンパク質の解析 多くの生物種でγ-チューブリン複合体が微小管のnucleationに重要であることが知られているが、線虫ではγ-チューブリン複合体構成因子の一部が見いだされておらず、その機能については不明な点が多い。本研究でγ-チューブリン(TBG-1)に結合するタンパク質を線虫抽出液から免疫沈降と質量分析で網羅的に同定した。そのなかから他の生物種のγ-チューブリン複合体構成因子と弱い相同性を示すタンパク質2つを見いだし、GTAP-1,GTAP-2と命名した。これらの因子のRNAiによって、TBG-1の中心体へのリクルートが減少することが見いだされたが生育には異常がなかった。このことはGTAP-1,2がγ-チューブリン複合体の構成因子であるが微小管形成能における役割は補助的であることを示唆している。
