Research Abstract

本研究では、生体分子を直接可視化でき、蛍光プローブ無しでも生体分子の機能分析が可能なナノ顕微観察技術の開発を目指す。生体の顕微観察においては、蛍光顕微鏡は必須のツールとして用いられてきたが、そこで実際に観察されているのは蛍光分子であり、生体分子ではない。染色せずに細胞や生体組織を顕微観察できれば、有害な色素を用いることなくより望ましい状態での生体機能を可視化できる。本研究ではエバネッセント照明と表面増強ラマン散乱を利用して、生体分子からのラマン散乱を高感度、かつ高空間分解にイメージングする技術を開発する。ラマン散乱は分子振動や配向の情報を光の波長、偏光、強度として持つため、蛍光色素に頼らずとも試料の分子そのものが観察像を与える。さらに金属表面から10nm程度の局所空間における局在プラズモンと生体分子と光との相互作用を利用して、ナノ空間における生体分子の機能を染色無しで把握することを目指す。
