Outline of Research at the Start

網膜疾患に対する再生医療は、社会的にも学術的にも大きな期待が寄せられているが、中枢神経系である網膜は、複雑な回路の形成過程や可塑的性質を有し、構造的にも機能的にも再生は容易ではない。網膜再生技術の確立には、正常・病変・再生網膜のいずれに対しても機能・機能不全の仕組みを明らかにし、網膜の設計図となる構成論的理解が必要不可欠である。本研究は、網膜回路動態の神経科学的解析および網膜再生技術の開発と連携し、網膜回路特有の動的特性が実現する情報処理機構および機能構築に関する数理モデルの構築を目指す研究である。

Outline of Annual Research Achievements

病変網膜活動のメカニズムの解析：視細胞が変性することにより発症する網膜色素変性症において、網膜の出力細胞である神経節細胞に観測される周期的自発発火のメカニズムを理解するため、網膜色素変性と同様の動態を示す、TrpM1KOマウス網膜を用い、組織解析およびパッチクランプ解析を行った。組織解析により、Trpm1KOマウス網膜では、桿体双極細胞（rod bipolar cell; RBC）からAIIアマクリン細胞へのシナプスの終末が縮退していることが知られているが、網膜変性の代表的モデル動物であるrd1マウスの網膜でも同様にRBC-AIIACシナプス終末の縮退が確認された。一方で視細胞-双極細胞の信号伝達を担うタンパク質であるmGluR6をノックアウトしたmGluR6KOマウス網膜では、そのような違いが見られなかった。rd1マウス網膜とTrpm1KOマウス網膜では、病変振動活動が見られるのに対し、mGluR6KOマウス網膜では見られない。前者２つでは視細胞-双極細胞の信号伝達が異なるにもかかわらず、同様の活動動態が見られることからRBC-AIIACシナプス終末の縮退が病変振動活動の主因子と考えられた。また前年度に構築した網膜数理モデルを用い、rd1、Trpm1KO、mGluR6KOマウス網膜のそれぞれの状態を想定したモデル改変を行いシミュレーションした結果、生理実験を再現できることが確認された。以上より、変性部位・過程の異なる様々な病変網膜において観測される病変振動活動は、共通のメカニズムを有することが示唆され、また構築した数理モデルはそれらを再現し、網膜ダイナミクスの理解に寄与する汎用的なモデルに値すると期待できる。
