Outline of Research at the Start |

Outline of Final Research Achievements |
In this study, we investigated the analgesic mechanisms of KCNQ channel opening with behavioral pharmacology and electrophysiology. In the behavioral experiments, systemic or intrathecal administration of the KCNQ channel opener retigabine showed analgesic effects, which were antagonized by systemic or intrathecal administration of the KCNQ channel blocker XE-991. Therefore, it was suggested that the opening of KCNQ channels is essential for the analgesic effect of retigabine, and that the spinal cord is important as the site of action. In the electrophysiological analysis, we studied the effect of retigabine in the spinal cord, which revealed that retigabine strongly suppressed excitatory neurotransmission derived from the primary afferent A-fiber, contributing to pain relief.

Academic Significance and Societal Importance of the Research Achievements |
