Outline of Research at the Start

近年、抗生物質が効かない細菌 (薬剤耐性菌)が世界規模で増加している。河川水といった環境水は、薬剤耐性菌や薬剤耐性遺伝子の拡散に寄与していると言われているが、その詳細はわかっていない。本研究では、河川水から単離した薬剤耐性腸内細菌科細菌の全ゲノム配列を決定して、保菌株や臨床分離株と比較することで、河川水が保菌や感染の原因となる薬剤耐性菌や薬剤耐性遺伝子の拡散に寄与している、という仮説を検証する。

Outline of Final Research Achievements

The increase of antimicrobial-resistant bacteria is a global health concern. In the present study, we sequenced the genomes of carbapenem-resistant Enterobacterales (CRE) recovered from river and lake water. The results indicated that environmental CRE carry rare/novel carbapenemases, such as FRI, GES-5, IMI, SFC, and SFH-1.

Academic Significance and Societal Importance of the Research Achievements

本研究により、環境水中の薬剤耐性菌は、臨床現場では珍しい薬剤耐性遺伝子を保有することが判明した。これらの薬剤耐性遺伝子を保有した薬剤耐性菌が実際に人々に取り込まれ、アウトブレイクを引き起こすかは、今のところわかっておらず、今後の研究で明らかにしていく必要がある。しかし、本研究では臨床現場でも報告例のあるカルバペネマーゼと菌種の組み合わせが検出されたことから、今後注意して環境由来の薬剤耐性菌をモニタリングしていく必要があることが示唆された。
