Outline of Research at the Start

乳児期に形成される腸内細菌叢は、一生を通してヒトの健康状態に大きな影響を与える。申請者は、これまでの研究において、母乳栄養児の腸管内でビフィズス菌優勢な細菌叢(ビフィズスフローラ)が形成される仕組みを解明した。そこにはヒトとビフィズス菌の共進化が強く推察される。本研究の根源的な問いは「乳児腸管でビフィズス菌を増やすことに、ヒト(乳児)にとってのどんなメリットがあるのか?」であり、それを探索的手法を通して解明することである。すなわち、乳児糞便のオミクス解析を通してビフィズスフローラ形成の意義を化合物(細菌代謝物)レベルで理解し、以て人工乳開発につなげるものである。

Outline of Final Research Achievements

We elucidated the molecular mechanism underlying bifidobacteria-rich microbiota formation in breastfed infant gut by genetically and enzymatically analyzing bifidobacterial enzymes involved in human milk oligosaccharide assimilation. We also found that bifidobacteria secrete a large amount of aromatic lactic acids, especially indole lactic acid, in the medium. Indole lactic acid is known to play important roles in maintaining intestinal barrier function and immune homeostasis. In addition, we found that several species of butyric acid-producing Firmicutes can assimilate both plant-derived oligosaccharides and HMOs. The results might imply adaptive response of these species to infant diet in the weaning period.

Academic Significance and Societal Importance of the Research Achievements

本研究ではまず、乳児腸管におけるビフィズスフローラ形成の分子メカニズムを解明した。次に、ビフィズスフローラ形成の意義の解明に取り組み、乳児型ビフィズス菌には高いインドール乳酸産生能があることを見出し、その遺伝子を同定すると共に酵素解析を行った。インドール乳酸は腸管バリアや免疫に重要な役割を果たすことが示されている。また、一部の酪酸産生細菌が植物オリゴ糖のみならず母乳オリゴ糖資化能を有していることを見出した。以上の成果は、乳児期の菌叢形成機序と意義を知る上で重要な成果であり、調整乳開発にも多大な貢献をした(母乳オリゴ糖の調製乳添加の科学的基盤となった)。
