Outline of Research at the Start

近年、様々な細胞を利用して実際の臓器の持つ機能を再現できる3次元構造物であるオル ガノイド(Organoid)を用いた基礎研究、発生生物学、疾患モデルとしての応用が活発に行われている。しかし、未だに骨細胞の特性また骨形成を再現できる in vitro モデルは報告されていない。申請者は旋回培養法にてiPS 細胞を用いた骨オルガノイドの作製法を確立し、実験管内での骨形成及び骨系疾患に対する薬剤スクリーニング、また骨疾患 iPS 細胞 を用いた in vitro 疾患モデルとしての活用が期待される。さらに骨オルガノイドを骨形成不全症モデルに移植し、再生医療への有用性を明確にする 。

Outline of Final Research Achievements

In this study, we fabricated three types of 3D scaffold-free tissue reconstructed by mouse pre-osteoblast cells (MC3T3-E1); 1) size-controlled spheroid under a stationary culture, 2) mass-produced spheroids under a rotational culture, and 3) cell disc using glass ring. Compared to a conventional 2D monolayer model, the 3D culture models elicited up-regulations of osteocyte markers in a short time even without addition of chemical osteogenesis supplements. The 3D culture models reconstructed by pre-osteoblast cells were scaled-up for further transplantation experiment toward bone regenerative medicine.

Academic Significance and Societal Importance of the Research Achievements

これまでゲルなどの人工物質を用いた様々な3次元培養法が報告されてきたが、これらの人工物質による移植後の拒否反応のリスクがあるのに対して、本研究では組織工学的手法を用いて細胞のみで構築される3次元培養モデルを作製した。さらに、前駆骨芽細胞の3次元化により骨細胞分化が短時間で誘導されることが明らかになった。本研究で作製された骨芽細胞由来3次元培養モデルは今後骨細胞のin vitroモデルとしての活用が期待され、さらに本研究により移植実験に応用可能なレベルまで3次元細胞組織体をスケールアップできた。
