Outline of Research at the Start

Bovine babesiosis caused by Babesia species is a serious threat to cattle industry worldwide. Our recent study demonstrated that Babesia sp. Mymensingh causes severe clinical babesiosis in cattle. However, unavailability of validated diagnostic tools and lack of knowledge on host range and global distribution undermine the control strategies. Therefore, in the present study, we will 1) validate a Babesia sp. Mymensingh-specific PCR assay, 2) identify the host range, and 3) determine the global distribution.

Outline of Annual Research Achievements

牛バベシア病は、経済的被害の大きい家畜の感染症で、アジアではBabesia bovis、Babesia bigemina、及びBabesia sp. Mymensinghの感染によって引き起こされる。そこで、モンゴルの21州のうち16州で飼育されていた計725頭の牛の血液DNAサンプルに対して、それぞれのバベシア種を標的としたPCRスクリーニングを行った。 そこ結果、B. bovis、B. bigemina、及びBabesia sp. Mymensinghの全体陽性率が、それぞれ27.9%、23.6%、及び5.4%であることが明らかになった。これらの結果より、モンゴルにおける牛バベシアの深刻な汚染実態が示された。特に、モンゴルにおけるBabesia sp. Mymensinghの検出は初めての報告となった。本論文は、モンゴル(Institute of Veterinary Medicine, Mongolian University of Life Sciences)との国際共同研究の成果である。

Reason

2019年度では臨床症状を引き起こす新たな牛バベシア種(Babesia sp. Mymensingh)に対する特異的な遺伝子診断用PCR法を確立し、かつBabesia sp.
Mymensinghの国際分布と感染宿主を解明できた。2020年度ではモンゴル国のBabesia sp. Mymensinghの汚染実態を明らかにできた。当初予定していた達成目標のほとんどを2020年度までに解決できたことから、当初の計画以上に進展していると判断する。

Strategy for Future Research Activity

異なる国々に分布するBabesia sp. Mymensinghや、異なる宿主に感染するBabesia sp. Mymensinghの遺伝子解析を行い、それぞれの遺伝学的特性を明らかにする。また、その特性から、病原性、宿主性、国際分布に関与する遺伝子多型マーカーを明らかにするとともに、Babesia sp. Mymensinghの起原を探索する。
