Outline of Research at the Start |
糖尿病発症の一因として膵β細胞の減少が挙げられるがその機序は明らかにされていない。現在までにインスリン抵抗性モデルマウスの肝細胞から分泌されるSerpinB1が膵β細胞の増殖を促進することを報告しており、本研究ではインスリン抵抗性下においてSerpinB1が発現誘導されるメカニズムについて明らかにする。

Outline of Final Research Achievements |
Insulin resistance and decreased insulin secretion are considered to be important factors of developing diabetes. We have reported a proteinase inhibitor, SerpinB1, as a novel humoral factor, which is secreted from the liver and facilitates compensatory pancreatic beta cell proliferation under insulin resistance. In this study, we investigated the mechanism by which SerpinB1 expression is induced in hepatocytes. In the liver of the db/db and ob/ob mice which exhibits severe insulin resistance, the expression levels of SerpinB1 were significantly elevated. Furthermore, an inflammatory cytokine, interleukin-6 (IL-6), the levels of which were elevated in these mice, induced SerpinB1 expression through JAK/STAT3 pathway in the liver. These results suggested that IL-6/JAK/STAT3 pathway plays an important role on the compensatory pancreatic beta cell proliferation in the liver.

Academic Significance and Societal Importance of the Research Achievements |
糖尿病発症の原因としてインスリン抵抗性やインスリン分泌低下が挙げられる。後者の原因として、インスリンを分泌する膵β細胞の減少が報告されている。一方で糖尿病発症初期には代償性に膵β細胞が増加することも報告されており、その機序を明らかにすることにより糖尿病を治癒出来る可能性がある。本研究では肝臓から分泌され膵β細胞増殖を促進させるSerpinB1の発現上昇機構を明らかにした。この研究をより発展させることにより糖尿病で膵β細胞が減少した患者の膵β細胞量を回復し、新規の糖尿病治療法の開発につながると考えられる。
