Outline of Research at the Start

パーキンソン病に対して脳内に電極を埋め込み刺激をする治療法が有効であるが、脳外科手術を必要とする。ウイルスベクターによる神経活動ブロックと、超音波の収束照射を組み合わせて、1)パーキンソン病モデルサルにベクターを全身投与し、2)脳局所に超音波を照射することにより血液脳関門を開けベクターを取り込ませ、3)目的の抑制性受容体が発現した段階で受容体に作用する薬剤を全身投与することにより脳局所の神経活動をブロックする、という非侵襲的なパーキンソン病治療法の開発を目指す。

Outline of Annual Research Achievements

パーキンソン病サルの大脳基底核に介入し、治療することを計画している。2023年度は、ウイルスベクターを用いて大脳基底核の活動に介入を行うことにより症状を改善することを試みた。
正常ニホンザルの大脳皮質運動野を電気刺激しニューロン活動を記録、早い興奮と遅い興奮からなる2相性の反応によって視床下核を同定し、抑制性のDREADDを搭載したウイルスベクターを注入した。DREADDの発現を待って、リガンド(DCZ)を投与したところ、ヘミバリスムと思われる異常運動が引き起こされた。また視床下核のニューロン活動を記録すると、発射頻度が減少していた。これらのことから化学遺伝学的手法により、視床下核活動がブロックされたことがわかった。次にドーパミン神経毒であるMPTPを総頸動脈に投与し、パーキンソン病モデルを作製し、同様に化学遺伝学的手法により、視床下核をブロックした。現在までのところ、パーキンソン病症状の顕著な改善は観察されていない。
また、別の発想として、興奮性ニューロンから成り立っている視床下核を抑制性に変換し治療することも考えられる。正常ニホンザルの視床下核にGABA合成酵素を搭載したウイルスベクターを投与した。正常なサルでは、大脳皮質運動野を電気刺激すると、早い興奮、抑制、遅い興奮からなる3相性の応答が大脳基底核の出力核である淡蒼球内節で観察されるが、ウイルスベクターを投与したサルにおいては、視床下核を経由すると考えられる早い興奮と遅い興奮が減少する傾向が見られた。

Reason

ウイルスベクターを用いた実験を開始するなど、実験計画は順調に進んでいる。一方、スペインCINACから共同研究者が来所するなど、コロナによって遅れていた国際共同研究も遅れを取り戻しつつある。

Strategy for Future Research Activity

パーキンソン病モデルを用いて、化学遺伝学的手法による視床下核の活動ブロックや視床下核ニューロンのGABA作動性化を行い、症状が改善するのか調べる。
