Outline of Research at the Start

本研究課題は、英国リバプール大学の構造生物学研究チームと共同で病因タンパク質の構造生物学的知見に基づいた筋萎縮性側索硬化症(ALS)の分子病態解明と新規治療法の開発を目指す。具体的に、2種類のALS原因タンパク質SOD1、TDP-43に着目し、SOD1については、その立体構造を安定化することにより疾患の進行を抑制する化合物の開発を行う。TDP-43については、ALSで見られる病理像の鍵となるTDP-43タンパク質の構造異常を英国チームと共同で解析する。本国際共同研究を通じて、新たな視点でのALSの病態解明、治療法開発や、異分野を俯瞰する国際的に視野の広い若手研究者の育成が期待される。

Outline of Final Research Achievements

Based on the structural analysis of Cu/Zn superoxide dismutase (SOD1), we synthesized a candidate compound that stabilizes SOD1 by expanding the structure of ebselen, a small molecule compound that binds to SOD1. We synthesized a candidate compound that stabilizes SOD1. Through screening ebselen analogues, we demonstrated that these compounds have neuroprotective effects against toxicity induced by mutant proteins in cultured neurons and amyotrophic lateral sclerosis (ALS) mice. Through this project, researchers in structural biology and neuroscience from the U.K. and Japan exchanged academic ideas and published the results in an international joint research paper.

Academic Significance and Societal Importance of the Research Achievements

ALS等の神経変性疾患では疾患に特徴的な病因タンパク質の構造異常が神経変性に深く関与するため、疾患・構造生物学の融合研究が不可欠といえる。本研究課題は、英日研究者の国際共同研究により病態解明を進め、学術論文として成果を発信した。学術的意義として、異分野連携による新たな知見創出と研究手法の確立が挙げられる。社会的意義では、難治性疾患克服への基盤構築と若手研究者の国際ネットワーク形成により、将来的な治療法開発と研究人材育成に貢献した。国際協力による包括的アプローチは、神経変性疾患研究に新たな展開を切り開くものである。
