Outline of Research at the Start

多能性幹細胞から得られる心筋細胞(PSC-CMs)は再生医療に加えて、疾患研究や創薬への利用が期待されている。しかし、PSC-CMsは、成体の心筋細胞とはその特性に大きな乖離がある。今後の応用を進めるためには、ヒト心筋細胞の成熟過程の理解と、その理解に基づいた成熟法や解析基盤が必要となる。Johns Hopkins大学Chulan Kwon准教授は新生仔ラットの心臓へPSC-CMsを移植することで、成熟した成体型のPSC-CMsを得る生体内成熟法を確立した。本研究では、生体内成熟法を用いることで、成熟過程のトランスクリプトームと、成熟レポーター細胞の開発と検証を行う。

Outline of Annual Research Achievements

本研究では、心筋細胞の成熟メカニズムの解明に向けて、研究を進めている。
コロナ禍の状況が落ち着いた2023年度は研究開始以来初めてJohns Hopkins大学への訪問が実現し、今後の共同研究の進め方についての打ち合わせを行うことができた。Kwon研究室メンバー全員との打ち合わせを行ったが、研究開始当初予定していた心臓内移植法の技術を持つメンバーが学位取得に伴い研究室を離れること、またその技術継承がうまく行っておらず現時点ではiPS細胞由来心筋細胞の心臓内移植が困難であることが明らかとなった。そのため今後の方針として、胚盤胞補完法、子宮内移植法など新しい手法の開発も含めて検討を行うこととした。
さらに、魚崎Gではin vitroでの成熟法の研究を進めており、一定の成果が得られており、in vitro側からのアプローチについても進めている。

Reason

iPS細胞由来心筋細胞を心臓内へと移植することで成熟させ、リファレンスデータを取る予定であったが、この実験を担当する予定であったKwon研究室メンバーが研究室を離れたことで、実験ができなくなっているため。

Strategy for Future Research Activity

新しい心筋細胞の移植法の開発、in vitroからのアプローチなどを進める予定である。
