Research Abstract

研究代表者らは生後の精子幹細胞の長期培養系(Germline Stem ; GS細胞)を確立し、この培養中にEmbryonic stem(ES)細胞と同様な奇形腫形成能と多分化能を持つ細胞が出現することを見いだした。本研究では奇形腫とseminomaの発症の分子メカニズムを明らかにすることを目的として行った。申請者らはGS細胞の腫瘍化の誘因の違いが、奇形腫とseminomaの違いを生むのではないかと考え、GS細胞の試験管内での形質転換を試み、多能性獲得につながる腫瘍化シグナルの検索を行った。
精子幹細胞株GS細胞ヘレンチウイルスによる発現ベクターを導入し、精子幹細胞からの奇形腫誘導に関わる遺伝子のスクリーニングを行った。Ras, Myc, Srcなど旧知のoncogeneや、Oct-4とNanog, Sox-2など多能性制御の関連遺伝子群、細胞周期調節因子のcyclinやCDK Inhibitorなどについて強制発現やDominant negative体を導入し、試験管内における増殖や形態変化、移植後の精巣における精子形成・腫瘍形成の有無を調べた。奇形腫の形成や試験管内にて形質転換を示すものは無かったが、cyclinのうち、Cyclin D2を高発現させた細胞は自己複製因子GDNF欠損下でも自己複製増殖し、移植後in vivoにてseminomaを作り、継代移植によりさらに腫瘍形成することから、Cyclin D2の発現は精子幹細胞の自己複製増殖に関わること、またその過度な活性化は癌幹細胞の発生を促すことが分かった。
