気管支喘息の全ゲノム関連解析:基盤構築と情報公開

Research Abstract

小児アトピー性喘息患者942名vs成人コントロール2370名の55万SNPの全ゲノム関連解析を行った。55万SNPのうち、常染色体に存在し、かつminor allele frequencyが0.01以上のSNPは453,047個であった。さらに、コントロールにおけるHardy Weinberg平衡がP<0.0001である1,170SNPを除外し、最終的な解析データとした。一次解析におけるGenomic Inflation factorは1.053であった。アレル頻度の比較では、6番染色体短腕のHLA領域で強い関連が認められた(P=7.7×10-9)。そのほかにも2番染色領域、12番染色体領域にP<1×10-7が存在した。HLA領域のSNPと小児アトピー性喘息との関連については独立した症例対照サンプルおよび家系サンプルにおいても追認されている(P<0.05)
さらに健康成人76名を対象としてHuman 610Quadを使用してタイピングを行い、別の研究成果として得られているCD4陽性T細胞およびCD14陽性モノサイトの網羅的遺伝子発現データ(イルミナHuman Ref8)と遺伝子型データを統合したデータベースの構築を情報支援班の支援を受けて作成しており、今後公開予定としている。
