Research Abstract

中枢時計SCNから行動リズム発振機構への出力経路を明らかにする目的で、時計遺伝子per1発現とPER2蛋白を発光にてレポートするper1-lucトランスジェニックマウスとPER2::LUCノックインマウスを用い、2以下の2実験を行い、所定の成果を得た。
1.活動開始と終了を制御する振動体の局在:明暗(LD)12:12h下で繁殖飼育したマウスをLD18:6の長日条件、又はLD6:18の短日条件に3週間以上暴露した後、SCNの水平断切片をルシフェリン含有培地にて培養し、1時間露光の連続発光イメージを5日間撮像した。ピクセル毎の位相マップを作成した結果、SCN前外側に点灯前からper1発現が上昇する細胞群の存在が分かり、最前部の細胞群のリズムとは180度の位相差があった。一方、長日下のPER2リズムには、SCN前後でper1同様の傾向はあったが、位相差は数時間であり、前SCNに夜明けのピークをもつ細胞群は存在しなかった。同一細胞が内因性リズム発振と環境応答で2つの分子ループを使い分けている可能性が示唆された。
2.遺伝子発現in vivo計測と行動リズムを駆動する振動体の検索:光ファイバーを用い、無麻酔・無拘束状態でSCNからの発光活性を連続計測し、時計遺伝子発現と行動リズムを同一個体で比較した。連続暗黒で飼育中のマウスに30分の光照射にてリズムをシフトさせ、時計遺伝子発現と行動のリズム変位の移行期を比較したところ、SCNにも行動リズムに一致した移行期をもつ細胞群の存在が明らかとなり、行動を駆動する中枢がSCN内部にも存在する可能性が示唆された。また、発光活性と自発活動は短時間の変動を示し、相互相関では自発活動がper1-lucを6-8分先行していることが分かった。per1発現からルシフェラーゼ蛋白合成までには数時間の時間を要するので、この変化は活動に伴う血流変化、特に、酸素、ATP、基質の変動などの影響を反映していると考えられる。
