大脳皮質神経細胞の配置制御

Research Abstract

大脳皮質層形成において必須の役割を有することが遺伝子変異マウスの解析等によって明らかであるにも関わらず、その生物学的機能が長年不明であるリーリンの移動神経細胞に対する機能を明らかにすることを目指した。これまでに、発生期大脳皮質に異所的にリーリンを強制発現することによって細胞凝集が誘導されることを見いだしたので、その詳細な解析を引き続き進めた。まず、受容体に結合できないリーリンを点突然変異で作成し、そのin vivoでの異所的強制発現を行ったところ、凝集塊は形成されないことを確認した。すなわち、細胞凝集は確かにリーリンとその受容体の結合を介した現象であることがわかった。次に、異所的凝集塊内において、遅生まれ細胞が早生まれ細胞を乗り越えて凝集塊の中心近くに配置される現象の特異性を検証するため、リーリン及びGFP発現ベクターを胎生14日で導入し、その後胎生16日にRFP発現ベクターとともにDab1のshRNA発現ベクターを導入した。その結果、Dab1がノックダウンされた赤色細胞(遅生まれ細胞)は緑色細胞(早生まれ細胞)による凝集塊の周辺に留まり、中心近くに向かって進入することはできないことがわかった。そこでさらに2種の既知のリーリン受容体(ApoER2及びVLDLR)についても同様の実験を行ったところ、やはり受容体がノックダウンされた遅生まれ細胞は凝集塊の周辺に留まった。以上より、リーリンは特異的なシグナル経路を使って移動神経細胞の凝集及び"inside-out様式"の細胞の配置を引き起こすことがわかった。
