Research Abstract |
昨年度までの解析から、前脳特異的にCREB活性型変異体CREB_<DIEDML>あるいはCREB_<Y134F>を過剰発現するマウスでは、多くのタイプの学習・記憶課題において、海馬のCREB活性と正に相関して、長期記憶ばかりではなく、短期記憶(30分-2時間程度)も向上することが明らかになった。今年度は、これら変異型マウスにおける短期記憶と長期記憶向上のメカニズムを解析した。 学習の難易度が高くなると変異型マウスでは、長期記憶形成の向上は観察されるものの、短期記憶の向上は観察されなることが明らかになり、CREBの恒常的活性化により短期・長期記憶の両方の向上が導かれるものの、短期記憶と長期記憶は異なるメカニズムで制御されるものと考えられた。 続いて、CREB活性型変異体発現マウスでは、CREB標的遺伝子群の恒常的な高発現が短期記憶向上を導いたものと考察し、CREB標的遺伝子であるBDNFに着目し、海馬BDNFの発現レベルを解析した。その結果、短期記憶向上が顕著なトランスジェニックラインほどBDNF発現レベルが高いことが明らかとなった。そこで、BDNF発現制御を介したCREBによる記憶制御のメカニズムを解析するため、海馬にBDNFあるいはBDNF inhibitor(K252a)を注入して、短期及び長期記憶を解析した。その結果、BDNF投与により野生型及びCREB活性化変異マウスの両者で短期記憶の向上が観察されること、一方、K252a投与により、野生型及び変異型マウスの両者で短期記憶の障害が観察された。従って、CREB活性化によるBDNFの高発現が短期記憶の向上を導いたと判断した。 さらに、BDNF投与により、CREB活性化変異マウスにおける長期記憶形成がより向上することも示され、BDNFの発現レベルの上昇とCREB活性化による記憶固定化能力向上の相互効果により、長期記憶形成がさらに強化されるものと考えた。
