Research Abstract

非染色、非侵襲で、分化に伴う細胞間相互作用を経て組織形成に至る一連の過程を分子論的に理解することを可能とする新規手法の開発が望まれている。本研究では検出信号としてレイリー散乱に着目し、顕微広帯域レイリー散乱分光により試料の静的な分光情報を、顕微動的光散乱により試料の動的な分子拡散に関する情報を、高空間分解能で同時に得ることのできる装置の構築を目的とした。
Ti : Sapphireレーザーの基本波(800nm,160fs,80MHZ)をフォトニック結晶ファイバーに導入し得られたフェムト秒白色光を倒立顕微鏡に導き、対物レンズを通して試料表面に集光する。試料からの後方散乱光を同じ対物レンズで集め、ピンホールを通した後にCCD分光器とフォトダイオードに同時に導く。金ナノ粒子(粒径;60nm)を試料として、面方向に200nm、光軸方向に650nmとサブミクロンの空間分解能を実現し、450-800nmの領域で分光測定が可能であると確認した。動的光散乱計測部はバンドパスフィルターで波長を切り分けて検出し、カウンティングボードにより自己相関曲線を導出することができる。作製した上記装置を用い単一マクロファージ細胞の計測を試みたところ、試料高さの増大に伴い細胞の散乱像の面積が減少しており、三次元像が明確に得られた。レイリー散乱は屈折率差が生じている箇所で発生するため、像はすなわち細胞内における空間ごとの分子濃度勾配を反映していると理解でき、スペクトルはその箇所に存在する分子群の電子スペクトルに対応する。本装置は生細胞イメージングに有効な計測装置であると確認した。
