Research Abstract

血管ネットワークは成体の隅々にまで張り巡らされ、酸素や栄養補給など重要な生理機能を発揮する。これらの血管ネットワークはどのようにして形成されるのであろうか?この問いに答えるために、初期胚における血管形成のしくみを研究した。個体発生の過程において、血管組織は一定のルールに従って作られる。そこでは動脈・静脈などのサブタイプに加えて、血管が作られるべき場所が厳密に規定される。血管形成の場所やタイミングがどのように制御されるのかについて、ケモカインSDF1とその受容体CXCR4の役割に注目した解析を行った。これまでに、SDF1/CXCR4シグナルは、背側大動脈から分岐する肋間血管の形成に重要な役割をもつことを見出している。興味深いことに、CXCR4は、形成されつつある細い血管において発現される一方で、それらの血管形成が終了し周囲を壁細胞で覆われる頃になると、CXCR4の発現が消失する。これらのことから、CXCR4は細胞移動の制御のみならず、血管内皮細胞の分化・成熟にも関わる可能性が見えてきた。これらの制御機構をさらに詳細に理解するために、CXCR4の下流で働く分子群の同定を試みた。候補分子の機能解析にあたり、トリ胚を用いた血管特異的な遺伝子導入法を開発した。解析の結果、さまざまな細胞接着関連分子群や、低分子量GTPase RhoファミリーがCXCR4と協調的に働く可能性が高まった。血管形成におけるケモカインシグナルの下流の分子メカニズムはこれまでほとんど知られておらず、本研究による成果は、血管パターンの形成機構を理解する上で、新たな知見を提供するものである。
