Research Abstract

ファンコニ貧血(FA)はまれな小児の遺伝性疾患で、臨床的には進行性の骨髄不全、高発がん性(白血病、扁平上皮がん)、骨格異常が特徴的である。患者由来の細胞はシスプラチンやマイトマイシンCなどのDNAクロスリンク薬剤に極度に感受性である。現在、13の原因遺伝子/相補群(FANCA、B、C等)が知られており、その産物はFA経路と呼ばれる共通の生化学経路で機能するものと考えられる。しかし、FA経路が実際に細胞内でどのようなDNAダメージを修復しているのかは、現在全く不明である。最近、正常細胞の代謝産物であり、血液中にも含まれているフォルムアルデヒドに対して、FA分子の欠損細胞(BRCA2/FANCD1と、FANCD2の欠損細胞)が特に高いフォルムアルデヒド感受性を示す事が報告された。本研究は、フォルムアルデヒドに対するFA分子の応答を解析し、メダカのFAモデルの作成を通じて、フォルムアルデヒドとFAとの関係を調べようとした。
1. メダカのFAモデルの作成のため、変異メダカライブラリーをいくつかの方法でスクリーニングを施行したが、残念ながら機能のロスが期待できる変異を同定できなかった。
2. フォルムアルデヒドのクエンチャー薬剤二種類添加のFA分子変異株の増殖への効果を調べた。FANCD2、FANCJ、FANCD1(=BRCA2)のいずれの細胞においても増殖スピードが加速されたが、一方野生型においてはその効果は全く見られなかった。FA経路分子がフォルムアルデヒドによるDNA損傷の除去に関与することが推定された。今後より詳細な解析が必要と思われる。
