Outline of Research at the Start

模倣と共感がどちらも生得的な知覚運動メカニズムとして捉えられるという指摘は早くからなされてきたが、これを実証しようとした研究は少ない。本研究は、小鳥の歌学習とラットの情動伝染をそれぞれ模倣と共感の原初的なモデルとして、神経回路から行動のレベルまで、それらの共通点を探ろうとするものである。仮説として、「模倣も共感も、ミラーニューロンと報酬系のドパミン神経細胞とが密に接続することで生起する」を提案する。本研究はコミュニケーション行動の生物心理学的基盤を与えることとなろう。

Outline of Annual Research Achievements

本研究では、模倣と共感の原始的モデルとして小鳥の歌学習とラットの情動伝染に共通する神経機構を探る試みを続けている。小鳥が歌を学ぶ過程では、他個体から聴き憶えた歌の記憶に基づいて自己の発声を修正する必要がある。この音声模倣学習に重要な機能を持つ可能性があるミラーニューロンとドパミン神経の関わりについて、神経解剖学的な研究を進めた。チロシン水酸化酵素はドパミンなどカテコルアミンの合成に必要な酵素である。Arcの遺伝子発現を可視化して同定された大脳基底核AreaXの聴覚発声ミラーニューロンに近接して、チロシン水酸化酵素が存在することを確認した。私たちはこの研究により鳥類の発声学習に必要な神経細胞に、ドパミンが直接投射している可能性を得た。ではこのミラーニューロンはどう形成されるのか。私たちは電気生理学的な手法により歌学習期の幼鳥２羽より神経活動記録を得た。結果、将来ミラーニューロンになるはずのニューロンは、学習初期では教示者の歌への聴覚応答のほうが強いことがわかった。まだ試験研究段階であるが、まずミラーニューロンは教示者の歌への選択性を獲得し、しかる後に自己の運動パタンを教示者の歌に近づくよう錬成するよう方向づける、という具体的な仮説をもって研究を進めることができよう。ラットを対象としたチロシン水酸化酵素の発現解析では、前部帯状回と偏桃体において発現を確認した。これらの部位には、情動発声を知覚する際と、情動事態に応答する際とに共に活動する広義の情動ミラーニューロンがあることが分かっている。情動ミラーニューロンをより簡潔に定義する方法と、そこに存在するドパミンがどう相互作用するかを示す必要がある。
