Outline of Research at the Start

本課題ではシングルセル解析を用いて腎臓構成細胞の亜集団を同定し、その機能を解明する。さらに、任意の細胞に任意の時点で細胞老化を誘導できる細胞老化誘導マウスを用いて、腎臓の各種構成細胞に細胞老化を誘導し、前述の亜集団の挙動や機能がどのように変化するかを検討するとともに、細胞老化の誘導が、周辺の細胞や臓器機能、障害応答性、さらには遠隔臓器にどのように影響するのかを解明する。

Outline of Annual Research Achievements

研究代表者は、腎臓の構成細胞に含まれる多彩な亜集団が重要な機能を担うことを見出し、当該亜集団の時空間的動態や制御機構の詳細な解析が必要と考えるに至った。ただし、未知の亜集団は従来の方法論では同定・分離・解析が不可能であるため、シングルセル解析を用いて、線維化、尿細管障害・修復、申請者らが見出した3次リンパ組織形成を含む、様々な腎臓病の素過程における腎臓構成細胞の亜集団を同定し、その機能を解明した。
一方で、研究代表者は、老化腎臓では若齢腎臓と比較して障害からの回復が悪いことに注目し解析してきたが、その差異が近年注目を集める「細胞老化」に起因するかは不明であった。本課題では、研究協力者の山田泰広教授との共同研究にて、任意の細胞に任意のタイミングで細胞老化を誘導できる「細胞老化誘導マウス」を用いて、細胞老化を腎臓の特定の細胞に時期特異的に誘導することの影響を個体レベルで解析し、臓器老化、ひいては個体老化との関連性を明らかにした。本課題では近位尿細管特異的、ポドサイト特異的、内皮細胞特異的誘導型Creマウスを用いることで、AKIで最も障害されやすい近位尿細管、増殖しない細胞であるポドサイトに細胞老化を誘導することで、前述の亜集団の機能がどのように変化するか検証するとともに、臓器老化指標、さらには障害からの可逆性がどのように変化するかを検討した。クレアチニンなどを用いた臓器機能評価に加えて、研究代表者は、前述の三次リンパ組織形成をふくめ、複数の臓器老化指標の数値化に成功していることから(未発表)、臓器老化の変化を定量的に評価した。

Reason

マウスの交配が順調に進み、表現型も明確なものが得られたため、当初の計画以上に進展した。
このプロジェクトの前に、細胞老化・臓器老化の評価指標が確立できていた点も、進展の一因である。

Strategy for Future Research Activity

今後は「課題1.シングルセル解析を用いた腎構成細胞の亜集団同定およびその機能解明」では、前年度までにマウスモデルで得られた知見を、カニクイザルモデルおよびヒトサンプルで検証したい。研究代表者は京都大学における新WPI拠点、ヒト生物学高等研究拠点(ASHBi)のメンバーである。本拠点はヒト、サル、マウスの種間比較を目的としており、研究代表者はカニクイザルのAKI-CKD移行モデルを確立している。本項目では、カニクイザルのAKI-CKD移行モデルでもシングルセル解析を行い、マウスで得られたデータと比較検討する。さらにカニクイザルで確認できた亜集団についてはヒト腎生検検体でもその存在を確認する。加えて、「課題2.遺伝子改変マウスを用いた臓器老化における細胞老化の意義の証明」では、前年度の解析をさらに発展させ、細胞老化に陥った細胞が障害・修復に与える影響を解明するとともに、周辺細胞や遠隔臓器に与える影響についても検証する。
