Outline of Research at the Start

磁性ナノ粒子を用いた新しい診断治療技術に関する研究課題である。腫瘍等に集積させた磁性ナノ粒子に体外から比較的低い周波数の交流磁界を印加し、そのときに生じる磁気信号を検出することにより体内の画像診断が可能となる。また、より高い周波数の交流磁界を印加すると磁性ナノ粒子が発熱する。この発熱は癌の温熱治療(ハイパーサーミア)に利用することができる。交流磁界に対する磁性ナノ粒子の磁化応答(ダイナミクス)を解明し、これら診断治療の実用を目指す。

Outline of Annual Research Achievements

1.磁性ナノ粒子の磁化ダイナミクス解明:10 ns~100 msという極めて広い時間領域で磁化ダイナミクスの計測に成功し、磁気緩和時間分布を解析した。高い温熱治療効果が得られるナノフラワー構造では、液中分散状態において、粒子の物理的回転が生じるにもかかわらず、粒子回転に由来する緩和時間が溶媒の粘性に比例しないという特異な傾向を観測した(Int. Conf. Sci. Clin. Appl. Magn. Carr. 2024 発表予定)。磁化ダイナミクスの高感度測定では、ネットワークアナライザとブリルアン散乱分光法を用いた独自のシステムにより、微小な磁化ダイナミクスの計測に成功した(Intermag2023)。
2.磁気粒子イメージング:頭部サイズでは交流励磁の頭部への影響を軽減できる弱磁界強度で動作させる装置(IEEE Trans. Magn. 2023)と、磁性ナノ粒子から十分な高調波信号成分を検出できる装置を完成させた。いずれも臨床診断で必要とされる1 μgの磁性ナノ粒子を検出可能なスペックである(IWMPI2024)。YBCO高温超電導線材を用いた傾斜磁界コイルを実装するボア径120 mmの人体1/5モデル装置を完成させた。磁性ナノ粒子のイメージングに成功し、空間分解能が向上することを示した。また人体全身サイズの臨床装置の実現において、一般的な銅コイルに対する超電導傾斜磁界コイルの優位性を定量的に明らかにした(Int. J. Magn. Imag. 2024)。
3.癌温熱治療:マウスに移植した腫瘍内に磁性ナノ粒子を投与して、磁化応答を計測した。緩和時間分布を解析することで、癌細胞種によっても異なる磁化応答が観測された。生きた腫瘍内における磁化応答のモデル化に成功し、癌温熱治療における効果的な粒子構造の解明に寄与する知見を得た(日本磁気学会2023年, 論文査読中)。

Reason

磁化ダイナミクスの高速測定及び広帯域測定を可能とする独自の計測システムとその測定手法を確立した。診断治療応用に使用される超常磁性を示す磁性ナノ粒子の磁化応答として粒子回転であるブラウン緩和、並びに磁化回転であるネール緩和を解明できた。いずれの緩和機構に対しても従来理論では説明できない現象を見出している。
磁気粒子イメージングでは、高温超電導コイルを組み込む磁気粒子イメージング実機を完成させ、磁性ナノ粒子の検出実験に移行した。人体全身サイズでの臨床装置を設計可能な唯一の仕様である。検出コイルとトランス接続する高感度磁気センサを利用する手法により頭部サイズでの磁性ナノ粒子の検出に成功しており、臨床診断に必要なスペックであることを検証した。
癌温熱治療への応用では、磁性ナノ粒子の発熱機構と発熱量の提示など当初計画は終了し、腫瘍内の磁性ナノ粒子の磁化応答の計測に成功するなど先駆的な成果を得ている。

Strategy for Future Research Activity

令和5年度の研究開始時点でも、残す2年間の研究計画を既に一部前倒しで実施してきた。令和5年度終了時点で全ての研究計画を実施完了、若しくは実施中の状況にある。申請時に想定していなかった従来理論に従わない磁化ダイナミクスの検証や、頭部サイズおよび人体全身サイズの1/5モデルの磁気粒子イメージング実機を用いた磁性ナノ粒子の検出、腫瘍内の磁性ナノ粒子の応答と発熱・イメージング特性との関係などを明らかにする課題を推進する。
