Outline of Research at the Start

天然物から有用な化合物が取得され、応用ならびに研究で利用されてきた。これらの化合物は表現型スクリーニングにより取得されているものも多く、分子標的が明らかにすることが重要である。しかし、化合物の標的分子同定は未だ難しい課題の一つであり、新たな手法の開発が求められている。本研究では、タンパク質が化合物と結合することによって熱安定性が変化することを利用したセルサーマルシフトアッセイとプロテオーム解析に基づくChemProteoBaseを組み合わせた新しい解析システムを開発し、新規化合物の標的を解析する。

Outline of Final Research Achievements

Target identification of bioactive compounds remains one of the most challenging tasks and has required the development of new methods. We developed and established a methodology for a new analytical system combining a cell thermal shift assay (CETSA), which uses the change in thermal stability of proteins when they bind to compounds, and ChemProteoBase, a proteomic database. We performed target analysis on bioactive compounds in the NPDepo library and on bioactive compounds isolated from natural sources with unknown targets.

Academic Significance and Societal Importance of the Research Achievements

生理活性物質の標的同定は、化合物の医薬や農薬などの応用開発における初期段階の重要なプロセスであるが、時間と手間のかかる過程である。本研究で開発した2DE―CETSAは生理活性物質の標的同定を迅速化する手法の一つになりうる。本手法は、現在のところ限られた細胞・限られた標的に対応するものであるが、他の技術との融合したり、さらに、この技術を発展させることにより、医薬などへの応用開発の貢献も期待される。
