Outline of Research at the Start

アミロイド線維や液-液相分離(Liquid-Liquid Phase Separation; LLPS)は様々な神経変性疾患と関連していると考えられており、その形成機構の解明は、アミロイド病の発症機構の解明や治療法の開発のために重要である。本研究では、多価の電荷をもつ核酸やATP、ポリリン酸などの生体高分子に焦点を当て、種々のアミロイド病原因蛋白質のアミロイド線維形成とLLPS形成の分子機構を解明する。これらの凝集体の生理的役割や構造的相関性を明らかにする。また、蛋白質の凝集体形成におけるポリリン酸類縁体の“促進と抑制”の相反する2つの作用機構について理解する。

Outline of Final Research Achievements

Using negatively charged biomolecules such as polyphosphate, the molecular mechanisms of amyloid fibril formation and protein aggregation due to liquid-liquid phase separation of α-synuclein, the protein responsible for Parkinson's disease, and β2-microglobulin, the protein responsible for dialysis amyloidosis were investigated.
Amyloid fibril formation was promoted by charge-charge interaction and preferential hydration with kosmotropic anions. Under the conditions of liquid-liquid phase separation, amyloid fibril formation was promoted by the depletion effect of biopolymers. Elucidating the mechanism of protein aggregation is important not only for the prevention and treatment of amyloid diseases, but also for understanding various physicochemical phenomena based on supersaturation.

Academic Significance and Societal Importance of the Research Achievements

本研究課題では塩やポリリン酸、ATPなど負電荷をもつアニオンに注目した。アニオンによる電荷-電荷相互作用と選択的水和、また、液滴界面における枯渇効果が、天然構造やアミロイド線維などコンパクトな構造を安定化することを明らかにした。これらの分子機構の解明は学術的に重要である。
また、過飽和は重要な物理化学的現象の一つであり、生体内では過飽和により蛋白質凝集が抑制されている。しかし、何らかのストレスにより、過飽和状態が崩れると、析出してアミロイド線維が形成される。アミロイド線維形成機構の解明は、過飽和の理解だけでなく、アミロイド病の予防や治療法の開発のために重要である。
