私たちは、大腸組織の解析から大腸上皮にエストロゲン受容体が発現しており，特に大腸がんの発生母地となる陰窩底部でその発現が非常に高いことを見出した。本研究では、私たちがこれまで作製してきた種々のエストロゲン欠損マウスモデルを用いて、エストロゲンと大腸発がんとの関連を明らかにする。またエストロゲンが，がん幹細胞の自己再生や分化，浸潤能，薬剤耐性能の獲得に及ぼす影響を大腸がんの手術検体を活用して明らかにする。マウスモデルおよび臨床検体を解析する本研究を通じて，女性の大腸がんの発がんの新機構を明らかにして，がんの予防および新規治療につながる基礎データを得る。

| Outline of Final Research Achievements |
The effect of estrogen on colorectal cancer progression was examined using cell lines, and we found that low levels of estrogen may inhibit colorectal cancer growth. While we were able to confirm the expression of estrogen receptor β in the crypts of mouse colon, we were unable to identify estrogen receptor α and endoplasmic reticulum membrane-localized estrogen receptor, suggesting that estrogen receptor β may play an important role in the development and growth of colorectal cancer.
The PDX study also supported this, but there were no significant differences among the mouse models. This study indicates that estrogen may be an aggravating factor for some colorectal cancers while acting in an inhibitory manner for colorectal cancer.

| Academic Significance and Societal Importance of the Research Achievements |
大腸がんは男性の病気というイメージが強いが，女性でも閉経後に大腸がんの発症率が上がる。しかも，発症部位にも違いがみられる。この違いにはエストロゲンが深く関与していると考えられているが，その機構はほとんどわかっていない。
我々の研究では閉経後女性や男性に発症する大腸がんとそこに関わるエストロゲンの関係を詳細に明らかにするため，モデルマウスを使って検討を行った。その結果，エストロゲンの濃度と大腸がんの増殖効率や発現遺伝子に変化が見られ，大腸がんの発症を抑えるにはエストロゲンの適切な濃度というのが存在すると分かった。
