申請者は紅色光合成細菌Rhodobacter sphaeroidesにおいて、細胞内炭素貯蔵物質として知られるポリヒドロキシ酪酸(PHB)が酢酸資化関連遺伝子の発現制御を媒介するシグナル分子として機能することを示唆する結果を得た。そこで本研究では、PHB蓄積を感知する転写制御因子PhaRの標的遺伝子をRNA-seqにより網羅的に探索し、PHBをシグナル分子とした代謝調節機構の全容を解明する。

Outline of Final Research Achievements |
By using an RNA-seq analysis, we searched genes upregulated by deletion of phaR encoding a polyhydroxybutyrate(PHB)-responsive transcription regulator. As a result, we found that genes involved in the ethylmalonyl-CoA pathway were upregulated by deletion of phaR, indicating that PhaR is involved in the regulation of acetate metabolism in R. sphaeroides. We also found that deletion of phaR and PHB synthase gene in the hydrogen-overproducing strain resulted in enhancement of hydrogen production and hydrogen yield from acetate

Academic Significance and Societal Importance of the Research Achievements |
本研究ではこれまで微生物において細胞内炭素貯蔵物質あるいはレドックスバランス調節物質として機能することが示されていたPHBが、代謝調節を媒介するシグナル分子としての役割を担うことを新たに示した。本研究成果はPHBをはじめとするポリヒドロキシアルカン酸生産菌の代謝生理を理解するうえで重要な知見であるとともに、ポリヒドロキシアルカン酸生産菌を対象とした代謝工学による物質生産菌の育種に資すものである。
