Outline of Research at the Start

世界の全ての大陸に400種以上が存在し,様々な有用植物を含むカキ属植物の用途は広く,さらに系統分類や進化学的にも興味が持たれてきた.本研究は,1)ゲノム情報と発現遺伝子情報の整備,2)機械計測と機械学習による形質評価法の開発,3)遺伝子機能評価系の開発,4)遺伝資源の調査収集の4種類のサブテーマの成果に支えられたカキ属植物の研究加速化のためのコモンプラットフォームを整備し,モデル植物の研究からは期待出来ない新発見や技術革新をもたらすために遂行するものである.これにより研究材料として最も扱いにくい木本性作物を研究対象にする果樹園芸学から基礎植物科学への貢献というパラダイムシフトの実例を示す.

Outline of Annual Research Achievements

本研究では,カキ属植物の基礎および応用研究を加速化するために,1)大量シークエンシング技術を活用したゲノム情報と発現遺伝子情報の整備,2)機械計測と機械学習による形質評価法の開発,3)遺伝子機能評価系の開発, 4)遺伝資源の調査と収集,の4種類のサブテーマにより支えられるコモンプラットフォームを整備するものである.本年度の成果は以下の通りである.
(ゲノム情報と発現遺伝子情報の整備)これまでに取得した10xGenomics社のGemCode Technologyを利用した六倍体カキゲノムデータに加え,本年度はPacbio HiFiデータも取得し,ハプロタイプレベルのゲノム構築に向けた情報解析を進めた.さらに,Iso-seqにより果実および葉の発現情報を収集した.加えて,カキの甘渋性遺伝子座のシークエンス解析を進めた.
(機械計測と機械学習による形質評価法の開発)レーザースキャンによりカキ果実のサイズと形状の三次元情報を数値化して評価する方法を検討し,カキ果実に存在する座や溝を評価する方法を確立して,論文発表した.また,当該計測手法を用いて発達解析を行い,カキに特徴的な溝形状の形成を制御する因子の有力な候補としてYABBYを同定した.また本年度は樹体生長を機械計測するための研究も,昨年度に引き続いて行った.
(遺伝子機能評価系の開発)昨年度までに開発したアグロバクテリウム法によるカキ属植物の形質転換系を活用して,カキ属植物の早期開花系統の作出に着手した.すなわち開花関連遺伝子であるCEN相同遺伝子を遺伝子編集およびRNAiによりノックアウトあるいはノックダウンするためのベクターを用いてカキの形質転換体を得た.
(遺伝資源の収集と整備)中国におけるカキの研究拠点のひとつである華中農業大学を訪問し,情報収集を行った.

Reason

COVID19の影響で,これまでほとんど実施できていなかった「遺伝資源の収集と整備」に関して,本年度は華中農業大学で調査を行うことができた点では,前進したと考えている.他の研究項目である「ゲノム情報と発現遺伝子情報の整備」,「機械計測と機械学習による形質評価法の開発」,および「遺伝子機能評価系の開発」に関しては,予定通りあるいは期待以上の成果が得られているのでおおむね順調な達成状況であると判断した.

Strategy for Future Research Activity

これまでCOVID19の影響を受け,当初の研究計画通りの研究が進んでいないカキの遺伝資源の収集と整備については,華中農業大学とならび中国におけるカキの重要研究拠点である西北農林科技大で開催される国際カキシンポジウムに出席し,情報収集を行う.あわせて他の研究項目である「ゲノム情報と発現遺伝子情報の整備」,「機械計測と機械学習による形質評価法の開発」,および「遺伝子機能評価系の開発」に関する研究を推進していく.
