Outline of Research at the Start

マイクロプラスチックの環境汚染が問題となる中、それらを捕集するための素材を、現在利用が進んでいない天然由来タンパク質を母体として調製することを目的とする。特に、タンパク質を高機能化するために感温性ペプチドで架橋することにより温度に依存して変化する網目構造を構築することを試みる。温度依存的に構造変化するタンパク質・エラスチンをモデルとして、エラスチン様ペプチドによる架橋反応をタンパク質に施しプラスチック粒子のサイズに対応した網目構造を構築することで、既存の石油化学製品とは異なる環境影響が非常に小さな、タンパク質性素材に結合する有害物質を除去可能で安全な吸着素材を開発することを目指す。

Outline of Annual Research Achievements

本研究では、天然由来タンパク質資源と新たにデザインした合成エラスチンペプチドアナログを用いた複合体を作製し、それらを用いてマイクロ粒子捕集素材、および、有害物質結合性素材の調製を行った。特に、天然エラスチン配列を改変した複数の新たな機能性エラスチンペプチドアナログの開発を行った。天然素材として、食肉産業からの畜産副産物としてブタ大動脈、また、海産物の副産物としてマグロ動脈球に焦点を当て、エラスチン(Elastin: E)やコラーゲン(Collagen: C)など様々な成分を含むタンパク質溶液(ECタンパク質溶液)の化学的、酵素化学的な効果的抽出方法の確立、抽出物の物性、抗酸化活性などの生理活性評価等を行なった。市販のタンパク質としては、BSA、パパイン、リゾチームなどを用いて、合成エラスチンペプチドを化学修飾タグとして結合し、生成したコンジュゲートの温度依存性、凝集性等の観測を実施した。前年作成した、BSAとエラスチンペプチドアナログ複合体を用いたマイクロプラスチックビーズの吸着試験においては、複合体の自己凝集能によりマイクロプラスチックビーズが溶液中から複合体の形成する沈殿とともに共沈殿することが確認された。また、本研究で調製した合成エラスチンペプチドアナログ・(FPGVG)5は、芳香族化合物と転移温度以下で相互作用し、アミロイド様構造の安定化によって自己集合能力を強化すること、さらに、集合体形成中に水溶液から環境化学物質として知られるビスフェノールAのような疎水性芳香族化合物を吸着・除去できることを示すという興味深い新しい知見を得た。本研究で得られたこれらの成果により、修飾分子としてのエラスチンペプチドアナログの有用性や市販および精製したタンパク質産物の機能化に資する知見を獲得した。
