Outline of Research at the Start

細胞の機械的な特徴とがん疾病の関係を明らかにするため、MEMSを駆使した細胞の機械特性計測と、その後のがん研究・検査に関連する生化学分析を実施する。基課題では、細胞の硬さと硬さに関連する遺伝子発現状態の関連性取得に取り組んでおり、これをがん研究応用へと進める。フランス北部でがん診断/治療技術の研究開発に取り組んでいるがん研究所、がん医療センターの研究者と協力し、機械特性結果と従来法によるがん検査結果との比較、クリニカルサンプルを用いた評価など行い、研究を発展させる。

Outline of Final Research Achievements

This research aims to reveal the cell mechanics of tumor diseases and plans to conduct characterization of single cells using MEMS, characterization of clinical samples, and comparing this method with conventional tumor tests. I could not have access to the clinical sample finally. Then, I characterized two cell lines in ONCOLille and continuously analyzed the gene expression level of Erbb2 on every single cell. Measured stiffnesses of breast cancer cells are softer than those of non-malignant breast cell lines, though the sample number is not enough to affirm it. The fluorescence signals at real-time PCR were not detected.

Academic Significance and Societal Importance of the Research Achievements

本研究の実施により、クリニカルサンプルではないものの、乳がん細胞と非悪性乳房細胞の機械特性について、がん化した細胞がより柔らかい傾向であることが示された。がんマーカーと機械特性との関連性評価について今後も研究を継続する予定である。複数のがんマーカーとの対応性など詳細な関係がわかれば、比較的短い時間で、がんの疑いのある浮遊細胞の検査を行うことも実現すると期待できる。
