Research Abstract

細胞の形態変化は,組織の構築から癌の浸潤,神経のネットワーク形成など多くの生命機能に深く関わる.特に,細胞膜下に発達するアクチン細胞骨格系はその主たる制御エンジンである.我々の先行研究において,アクチン線維の脱重合から放出されたアクチン単量体の細胞内濃度変動や密度不均一性が,mDia1や類縁のフォルミンファミリーといった重合促進分子による細胞内アクチン線維の形成活性を著明に促進させることが見出された.これは,アクチン線維の再生を促すフィードバック制御機構の存在を示唆している.本研究では,細胞骨格を障害する物理ストレスに対する応答時における,このアクチン脱重合-線維回生のフィードバック連関の役割について,細胞内分子可視化を用い検証した.その結果,mDia1は,細胞への物理刺激に迅速に応答し,10秒以内に活性化,その活性化には,アクチン重合活性部位だけで十分,mDia1のN末端にある制御配列は必要とせず,類似のフォルミンファミリー分子も同様の反応を示すこと、細胞内のカルシウムイオンやリン酸化酵素を阻害しても,この活性化は誘発されることなどを見出し,アクチン細胞骨格の物理的損傷を回復するための新たな分子機構の存在を解明した(論文投稿中).また,フォルミンファミリーはアクチン重合端にプロセッシブに結合しながら連続的にアクチン伸長を行うことを,2004年我々は直接可視化し証明したが,インビトロの1分子蛍光偏光観察によって,mDia1がアクチン線維の螺旋構造に沿って回転することを見出した.線維軸まわりの回転方向への力学的な影響がフォルミンファミリーによるアクチン重合と関わる可能性が示唆される成果である(2011年Science誌掲載).
