Research Abstract |
真核細胞のDNAは、H2A-H2B二量体とひとつのヒストン(H3-H4)_2四量体からなるヒストン八量体に巻き付き、ヌクレオソーム構造をとる。ヌクレオソーム構造は相同組換え反応を含めた様々なDNA介在反応を負に制御している。従って、相同組換えを利用した遺伝子のノックアウトの高率化において、組換え反応時のヌクレオソームの制御機構を理解し、その原理をノックアウトの高率化に応用することは重要である。申請者は、昨年度の実績としてヒストンのアミノ酸を個々にアラニンに置換した酵母の点突然変異株ライブラリー439株の解析を行ない、相同組換えに欠損がありそうな90株のMMS感受性点変異株を同定していた(Genes Cells 14,1271-1330,2009)。この中から、MMS感受性を示す、H3-K56A変異株に焦点あてた。H3-K56はアセチル化(H3-K56-Ac)されることが知られていたが、本研究でH3-K56-Acに依存して、相同組換えの一種である姉妹染色分体間の組換え(SCR)が起ることをはじめて見いだした(GEnes Cells 15,945-958,2010)。興味深いことに、DNAヘリカーゼSgslの欠損あるいはヒストンシャペロンCAF-1の欠損により、それぞれSCRが野生株に比べて亢進することを見いだした。さらにCAF-1とSgslの同時欠損酵母細胞ではSCRが相乗的に頻発した。これは、ヒト細胞でCAF-1とSgslの相同遺伝子を同時に低下させることで、ヒト細胞に高効率のノックアウトを実現できる可能性を示したものといえる。
