Outline of Research at the Start

本研究では、新しい水質センシング技術で安全性を確保する、塩素消毒とRO膜のみのシンプルな過疎地域用の小規模水道システムを提案する。病原体は医学分野でバイオマーカーの検出に活用されているDNAアプタマーで分析する。本水道システムは過疎地域のみならず、災害発生時の飲料水の安全性確保、途上国の井戸水の水質管理にも適用できる。新しい病原体センシング技術は人間や家畜の感染症の診断にも適用できる。

Outline of Final Research Achievements

A gold nanoparticle probe method was developed to detect the eae gene possessed by enterohemorrhagic Escherichia coli O157:H7. As the concentration of eae gene mRNA decreased, the intensity of scattered light also decreased. However, compared to AOB, the overall scattered light intensity tended to be higher. While the peak scattered light intensity at an eae gene mRNA concentration of 12 copies/μL was 148.1, the peak scattered light intensity at an AOB 16S rRNA concentration of 40 copies/μL was 39.8. These results suggest that the type of probe DNA used and the target nucleic acid can influence the intensity of scattered light.

Academic Significance and Societal Importance of the Research Achievements

腸管出血性大腸菌感染症の原因菌はベロ毒素を産生する大腸菌である。ベロ毒素は培養細胞 の一種であるベロ細胞に対して致死的に作用することからこの名前が付けられている。ヒトを発症させる菌数はわずか50個程度と考えられており二次感染が起きやすいのも少数の菌で感染が成立するためである。また、この菌は強い酸抵抗性を示し、胃酸の中でも生残する。本研究では、このようなヒトの健康を害する病原体の遺伝子を新規の方法で検出することに成功した。
