| Outline of Research at the Start |
本研究は出生コホートを基盤とし、環境化学物質曝露による健康影響の機序を明らかにすることを目的とする。保存済みの臍帯血、前向きに追跡している参加児の生体試料を用いて、有機フッ素化合物、フタル酸エステル類の曝露評価を行う。子どもの各種ホルモン、カテコールアミン類、炎症メディエーター等を測定し、曝露との関連、疾病との関連を疫学研究で解明する。動物実験では、曝露による典型的な遺伝子発現の変化を確認し、曝露マーカーとなり得るタンパク質あるいはホルモン等を探索する。最終的に、曝露によるAdverse Outcome Pathwayの概念に基づく評価を行い、曝露と健康との関連における機序を解明する。

| Outline of Final Research Achievements |
In this study, we aimed to elucidate the mechanisms underlying the association between environmental exposures and health outcomes by integrating human epidemiological research with animal experiments. In the human study, we analyzed blood and urine samples collected from prepubertal-aged children participating in the "Hokkaido Study" to measure levels of phthalate esters and organophosphate triesters metabolites, steroid hormones, type 2 biomarkers, cytokines, and short-chain fatty acids. We identified associations between exposure to these chemicals and the prevalence of allergic conditions, as well as correlations with the aforementioned biomarkers. In the experimental study, we investigated the PPARα agonist activity of PFOA and DEHP metabolites and examined gene expression in human cells. Furthermore, we identified candidate genes whose expression was altered in the livers of neonatal mice following exposure, suggesting potential mechanisms of toxicity.

| Academic Significance and Societal Importance of the Research Achievements |
本研究では前思春期の子どもの化学物質曝露濃度と、T2バイオマーカー、性・ステロイドホルモン、炎症性サイトカイン、単鎖脂肪酸などの様々なバイオマーカー値を明らかにした。フタル酸エステル類やリン酸トリエステル類曝露は、酸化ストレスを引き起こし、それがT2炎症反応を介してアレルギー症状につながる可能性を示唆した。また、性・ステロイドホルモン値のかく乱、二次性徴発来状況の遅延、INF-α等炎症性サイトカインの増加、単鎖脂肪酸の減少と関係する可能性を示唆した。特に重要なのは、これらの関連が小児の一般集団で観察されたことである。化学物質曝露から将来の世代の子どもの健康を守るための重要な知見を示した。
