Outline of Research at the Start

本研究は、食品因子による未病段階での認知症の改善・予防を実現することである。すなわち、認知機能低下を未病段階で予防・改善する食品因子を予測し、実践できる認知症予防食品科学を構築する。血液脳関門(BBB)を透過できる、腸管吸収性が保証される、標的受容体へ結合し、脳機能を改善できる食品因子を明示し、食生活からの認知症発症予防の可能性を明らかにする。

Outline of Final Research Achievements

This study aimed to elucidate the improvement and prevention of dementia at the pre-disease stage through food factors. A BBB-transportable peptide Tyr-Pro and other food factors were investigated for dementia prevention effect when taken orally. As a result, it was found that Tyr-Pro reaches the brain parenchyma after oral injection and improves the decline in spatial learning ability by in silico, in vitro, and in vivo studies. This was due to the activation of the acetylcholine system via AdipoR1 and the inhibitory effect on amyloid beta accumulation. We also clarified that the intake of lactic acid, ferulic acid, etc. in combination with exercise is effective in preventing dementia.

Academic Significance and Societal Importance of the Research Achievements

神経変性疾患である認知症のなかでも、アルツハイマー型認知症は発症数が増大し、大きな社会的解決課題の一つとなっている。治療薬としてレカネマブが上市されたものの、遅延薬であるため、根本解決にはやはり発症前の予防や改善が極めて重要である。本研究では、未病段階で改善作用を示す食品因子を明らかにした点で大いに評価され、認知症を未病段階で改善できる新たな機能性食品の開発に資する成果を得ているといえる。
