Outline of Research at the Start

植物には神経が無いが、動物の神経と同じように活動電位と呼ばれる電気信号が知られている。しかし、従来の研究材料では研究が難しく、どのように電気信号が発生し、伝わっていくかの仕組みはわかっていなかった。本研究では、活動電位を研究しやすい、食虫植物のモウセンゴケ、ハエトリソウ、ムジナモ、お辞儀運動をするオジギソウを用いて、活動電位発生伝搬の仕組みの基盤を明らかにするとともに、その進化を推定することを目的とする。

Outline of Annual Research Achievements

1)センサーラインの作成：モウセンゴケでpHusion、ClopHensorNラインが完成した。２）チャネルロドプシン解析：モウセンゴケにおいてACR1-eYFPとjRCaMP1a、XXM-eYFPとjRGECO1a、ハエトリソウについて、XXM-eYFPとjRGECO1aを導入した形質転換ラインを得た。３）遺伝子破壊：[モウセンゴケ]single cell transcriptome解析は核単離が困難だったため、触毛の頭部、柄、基部のRNA-seq解析を行った。顕微鏡観察に適したアントシアニン欠損（AF）野生型のゲノムリシーケンスを完了、AF野生型標準株とした。MSL10, CNGC15、GLR3.6、TPK1、AHA2a、AHA2の遺伝子破壊体を作出した。[ハエトリソウ]MSL10破壊株において、Ca2+動態を調べた結果、野生型より機械刺激感受性が低下していること、活動電位の発生確率が下がることがわかった。４）局在解析実験：細胞膜局在マーカーラインを確立した。自己プロモーター::遺伝子cDNA-mClover3遺伝子の形質転換実験を行っている。５）オジギソウの解析：MSL10、GLR、AS/LOB遺伝子破壊体などの解析から、MSL10とGLRが正常な活動電位とCa2+波の発生に必要であること、MSL10が機械刺激受容体として機能していることがわかった。さらに、AS/LOB遺伝子がMSL10とGLRを正に制御していることがわかった。６）電気生理学的解析、誘導実験系、相互作用因子解析、オルソログ遺伝子のシロイヌナズナでの機能解析、多様性解析：モウセンゴケ、ハエトリソウ、オジギソウにおいて、細胞内電極挿入による電気生理実験手法をほぼ確立し、Ca2+波と活動電位の同時測定を開始した。また、ハエトリソウにおいて、細胞破壊実験によって、活動電位発生細胞を特定することに成功した。
